It is impossible to dig up a plant planted inside a vine plant without first removing the vine plant themselves unless the vine is endangered.

Differences in the Chinese version[1]
While they are still able to be planted over other plants, they cannot be planted on their own. If a plant should be defeated or otherwise disappears from inside of a vine, such as Puff-shroom despawning, the vine will eventually despawn on its own unless another plant is placed inside.
Cob Cannon and P-Mech Assembler - Flamestar cannot have vines planted over them as they take up two tiles when planted, and Cob Cannon cannot be planted over any Kernel-pults with vines on them. Vines need Lily Pads and Flower Pots to be planted on water and roof tiles respectively, and cannot be planted on sky tiles at all.
When a zombie attacks a plant inside of a vine, the vine will be damaged first. Zombies that perform crushing attacks such as All-Star Zombies and Gargantuars will crush both the vine and the plant inside, with the exception case being Pumpkin, who will tank the damage individually. Intensive Carrot will only revive the plant inside the vine should both be defeated.
If a shovel is used on a tile occupied by a plant with a vine over it, both the plant and the vine will be dug up simultaneously. If a plant and a vine is planted on a Lily Pad or Flower Pot, the plant and the vine will be dug up simultaneously, but the Lily Pad and Flower Pot will remain.
Vines are not counted towards the objectives of levels that count the number of plants (e.g. "Do not have X amount of plants", "Do not lose more than X amount of plants"), nor do they count for Two-Player Mode's plant limit.
If a vine is wearing a Pendant, the effects of the pendant will not visually appear, but will still take place. Vines can be planted on water on their own if they're wearing a Ducky Tube Pendant, but cannot be planted over other plants wearing Ducky Tube Pendants.
Trivia
- Despite functioning as his family's equivalent to one, Pumpkin is not considered a vine plant, as he is not present in Store sales involving them and his official Reddit reveal post only refers to Pumpkin as being "similar to Vines".
- This is however, not the case in the Chinese version of Plants vs. Zombies 2, where Pumpkin is explicitly referred to and categorized as a vine plant.
- Glitter Zombie can kill Power Mints that have been planted on vine plants.
Specific to the Chinese version of Plants vs. Zombies 2
- As confirmed by Pumpkin's Chinese reveal post[1], the in-universe reason for why vine plants cannot be planted by themselves is that they derive nutrients from the plant they are surrounding.
